چکیده

Novel biosurfactant-producing strains were isolated from hydrocarboncontaminated environments that exclusively utilize agro-waste as their primary carbon source for the expression of biosurfactants. These were quantified using various standardized methods. Among the agro-waste screened, Beta vulgaris (Beetroot) proved to be the most suitable substrate, for which the biosurfactants produced by three bacterial isolates–B. licheniformis STK01, B. subtilis STK02, and P. aeruginosa STK03–lowered the surface tension of the culture media to 30.0, 32.98, and 30.37 mN/m, respectively. The biosurfactants achieved considerable emulsification activity, particularly for heavy hydrocarbons, with the highest emulsification indices being 65.5% and 95% for anthracene and lubricant oil, respectively. The emulsion formed with lubricant oil was thermally stable even up to 50 °C for 21 days. The results showed the proficiency of the novel bacterial isolates used, as well as the suitability of solid agro-waste for biosurfactant production, thus suggesting that exclusive utilization of solid agro-waste is a promising option for use in biosurfactant production for environmental remediation. The outstanding emulsification activity and thermal stability demonstrated by the biosurfactants produced showed their potential applications in enhancing bioavailability and bioremediation of recalcitrant and hydrophobic environmental contaminants.

Assessment of different screening methods for selecting biosurfactant producing marine bacteria

Different screening methods namely, hemolytic assay (HA), modified drop collapse (MDC), tilted glass slide (TGST), oil spread method (OSM), blue agar plate (BAP), hydrocarbon overlaid agar (HOA) plate, emulsification index (EI), emulsification assay (EA) were assessed for their efficiency to detect biosurfactant producing marine bacteria. Forty-five strains of bacteria, comprising 18 Acinetobac...

Diversity of Biosurfactant-producing Streptomyces Isolates from Hydrocarbon-contaminated Soil

Actinomycetes are unsurpassed as producers of bioactive metabolites, primarily those with antimicrobial/anticancer properties; however, their capacity for producing other natural products such as biosurfactants, has been less explored. Biosurfactants are heterogeneous surface active compounds and can be potentially deployed for bioremediation of hydrocarbons and as emulsifying agents in agricul...

Biosurfactant production by a newly isolated soft coral-associated marine Bacillus sp.E34: Statistical optimization and characterization

Marine biosurfactant-producing bacteria were isolated from the soft coral Sarchophyton glaucum collected from Red sea. The main criteria used for screening of biosurfactant producers were haemolytic activity, dropcollapse, oil displacement and emulsification index. Based on phenotypic characterization and analysis of 16S rDNA sequencing the most potent isolate was identified as Bacillus sp.E34....

Evaluation of Screening Methods for the Isolation of Biosurfactant Producing Marine Bacteria

Hemolytic assay, bacterial adhesion to hydrocarbons (BATH) assay, drop collapse assay, oil spreading assay, emulsification assay and surface tension measurement were compared for their reliability and ease of use to screen biosurfactant producing bacteria. 105 bacterial strains were screened for their biosurfactant production. Results from screening methods revealed that hemolytic and BATH assa...

Screening and Identification of Potential Biosurfactant Producing Microorganisms Isolated from Oil Contaminated Soils

Biosurfactant are surface active compounds produced by microorganisms. These molecules reduce surface tension between aqueous solutions and hydrocarbon mixtures. Hundred hydrocarbon saturated soil samples were collected from thirty different areas in Chennai, TamilNadu, India. Seventy two bacterial strains were isolated and cultured by enriching carbon (Glycerol) source. Each culture medium was...
